Four Substantially Proven Strategies for Our Longer Health and Lifespan

Exploring the Science Behind Supplementation for Longevity

In the quest for longevity, four supplement strategies have emerged with substantial scientific backing: raising NAD+ levels, using senolytics to eliminate senescent cells, supplementing with Fatty 15 (C15,0) and utilizing low-dose lithium. Below, each strategy is supported by data from recent studies. I have appended my personal experience with each of these strategies for consideration.

Raising NAD+ to Youthful Levels

Several studies have highlighted the anti-aging potential of boosting NAD+ levels. In a 2016 Cell Metabolism publication, mice supplemented with nicotinamide mononucleotide (NMN) showed enhanced mitochondrial function, increased energy, and improved insulin sensitivity.

Human trials, such as a 2020 double-blind, placebo-controlled study published in Scientific Reports, found that NR supplementation increased NAD+ levels in healthy adults by up to 60% in four weeks, with improvements in markers of cardiovascular health. These results suggest that restoring NAD+ could play a vital role in staving off age-related decline and promoting cellular health.

A 2023 review article published in Drugs & Aging highlights the importance of NAD+ anabolism in metabolic, cardiovascular, and neurodegenerative disorders. The study emphasizes that raising intracellular NAD+ levels represents a promising strategy to improve these conditions.

Senolytics to Eliminate Senescent Cells

Evidence for senolytics comes from both animal and preliminary human studies. In a pivotal 2018 Nature Medicine study, mice treated with the combination of dasatinib and quercetin experienced a significant reduction in senescent cell burden, improved physical function, and prolonged healthspan.

More recently, a 2019 pilot human trial published in EBioMedicine involved patients with idiopathic pulmonary fibrosis; after receiving oral senolytics, they demonstrated improved physical performance and a decrease in markers of cellular senescence. These findings indicate that targeting senescent cells may slow aging and enhance tissue health.

A 2025 review in Biomolecules discusses recent advances in senotherapeutics, focusing on senolytics and senomorphics for anti-aging interventions. The study highlights the potential of these strategies to target senescent cells and modulate the senescence-associated secretory phenotype (SASP).

I have used Senolytic Activator from Life Extension for nearly 5 years, at first quarterly, now every 2 months. LE uses Theaflavins in place of the drug Dasatinib, which engages the same receptors, causing cell death in senescent cells. I use this in a Hit-and-Run strategy: I take a bottle of 36 capsules in 3 days; 4 caps 3x/day on an empty stomach, then wait 2 months. The first time I did this I had flu-like symptoms on day 3, stopped for a day, then resumed. I have had no symptoms since, which I believe means very low accumulation of senescent cells after my years of use.

Fatty 15

Fatty 15 is based on research into the saturated fatty acid C15:0. According to a 2022 study in PLOS ONE, supplementing with C15:0 improved metabolic and inflammatory markers in marine mammals, and subsequent pilot human studies suggested similar benefits. The research team found that participants taking Fatty 15 for 12 weeks exhibited lower levels of systemic inflammation, improved cholesterol profiles, and enhanced cellular membrane stability. The results, though early, point to Fatty 15 as a promising dietary addition for heart and metabolic health.

Additionally, a 2025 article by Dr. Venn-Watson further validates C15:0 as an essential fatty acid, emphasizing its role in maintaining long-term metabolic, heart, liver, and red blood cell health. In the Navy Dolphins they work with, highest C15:0 tissue levels yield upwards of two decades of increased healthy life. In the wild dolphins live 20 to 25 years; the NAVY has some approaching 60.

Low Dose Lithium

The longevity effects of low-dose lithium are supported by population studies and laboratory research. A landmark 2011 study published in the European Journal of Nutrition analyzed drinking water lithium concentrations across 18 Japanese municipalities and found that communities exposed to higher trace lithium experienced lower standardized mortality ratios. In animal studies, low-dose lithium extended lifespan in C. elegans and provided neuroprotective benefits in rodents.

Additionally, a 2014 review in Translational Psychiatry emphasized lithium’s potential to reduce dementia risk at microdoses far below psychiatric levels, suggesting a meaningful role in aging and cognitive preservation.

I have taken 5 mg/day for the last 30+ years; my wife still says I remember nothing!

These supplement strategies are supported by growing evidence, but individuals should consult healthcare professionals (if you have one that cares – mine never have a clue) before considering new regimens. Collectively, they represent the vanguard of modern longevity science, promising a future of healthier, longer lives.
